Ralph has starred in a lot of films during the course of her career. She won the Independent Spirit Award for Best Female Supporting Actress in 1991 for her performance in the drama film To Sleep with Anger. Among her many film credits are The Mighty Quinn (1989), The Distinguished Gentleman (1992), Sister Act 2: Back in the Habit (1993), and Deterrence (1993). (1999). Ralph featured in the syndicated comedy It's a Living from 1986 to 1989 and then starred in the short-lived ABC sitcom New Attitude (1990) and CBS medical drama Street Gear (1990). (1995). She played as Dee Mitchell in the UPN comedy Moesha from 1996 to 2001 and as Maggie Turner in the Nick at Nite series Instant Mom from 2013 to 2015. Sheryl Lee Ralph's first book, Redefining Diva: Life Lessons from the Original Dreamgirl, was published by Simon & Schuster in 2012. She made her Broadway debut in 2016 as Madame Morrible in Wicked.
